![]() It was a time of war, of the breaking up of Roman Britannia into several separate kingdoms, of religious conversion and, after the 790s, of continual battles against a new set of invaders: the Vikings.Ĭlimate change had an influence on the movement of the Anglo-Saxon invaders to Britain: in the centuries after 400 AD Europe's average temperature was 1☌ warmer than we have today, and in Britain grapes could be grown as far north as Tyneside. However, most historians now prefer the terms 'early middle ages' or 'early medieval period'. The period used to be known as the Dark Ages, mainly because written sources for the early years of Saxon invasion are scarce. The Anglo-Saxon period in Britain spans approximately the six centuries from 410-1066AD. We hereafter refer to this model as the liver-specific double-knockout (L-dKO) mouse. We isolated tumors from an HCC mouse model generated by liver-specific deletion of Pten and Tsc1 (Fig. These mediators are potential, novel drug targets. While, some of the mediators are known oncogenes, others are novel oncogenic mediators. The identified mediators include transcription factors, kinases, phosphatases and deacetylases. NetICS employs a sample-specific network diffusion process that reveals the convergence of diverse changes in distinct tumors (mutations and differentially expressed miRNAs) on common downstream mediators. NetICS provides a comprehensive framework that reveals how specific genetic aberrations (i.e., deletion of the tumor suppressors Pten and Tsc1) and tumor-specific changes in miRNA expression can affect downstream mediators. In this study we use NetICS, a computational method to integrate multi-omic data (somatic mutations, miRNA differential expression, transcriptomics, proteomics and phospho proteomics) from an mTOR-driven mouse HCC tumor model, to understand the molecular mechanisms of mTOR-driven HCC. New methods are necessary to integrate different types of omics data to identify dysregulated pathways. However, a major drawback of these studies is that they rely solely on genomic analysis. Computational methods that focus on the direct effect of genetic aberrations, i.e., the effect of a gene mutation on the encoded protein, have also been proposed. For example, integration of DNA, RNA and phosphoproteomic data enabled stratification of prostate cancer patients and to identify individualized treatment options. Recently, multi-omics analysis has been informative in the characterization of tumors. To identify such mediators, it is essential to integrate omics data, i.e., the genome, transcriptome, proteome and phosphoproteome (commonly referred to as multi-omics analysis), from diverse tumors. Distinct genomic aberrations (in different tumors) are expected to converge functionally on the same downstream protein, referred to here as a ‘mediator’. However, to further understand the effect of a genetic aberration or dysregulated gene expression (possibly due to aberrant miRNA expression) it is necessary to identify the mediators common to diverse alterations. Transcriptomic and proteomic analyses have been performed to stratify HCC patients into clinically-relevant groups. Common mechanisms that can dysregulate miRNA expression in human cancers include amplification, deletion or epigenetic changes. miRNAs are key regulators of the transcriptome and can act as either oncogenes or tumor suppressors. Apart from somatic mutations, tumorigenesis can be regulated by the levels of specific miRNAs, mRNAs, proteins and protein phosphorylation. In addition, different mutations across cancer patients (genetic divergence) could result in the same pathways being activated (functional convergence). A limitation of relying entirely on genomic data to determine a therapeutic strategy is that it ignores functionally-related, non-mutated genes that could also encode potential drug targets. While DNA sequencing has enabled a comprehensive characterization of tumor genomes and stratification of patients, translating such information into treatment strategies has remained a major challenge. We generated an mTOR-driven HCC mouse model, by liver-specific deletion of the tumor suppressors Pten and Tsc1, to investigate the molecular and cellular mechanisms of mTOR-driven tumorgenicity. However, the effector pathways via which mTOR promotes tumorgenicity are widely unknown. Approximately 50% of HCC tumors exhibit loss of the tumor suppressors Pten, Tsc1, or Tsc2 leading to aberrant PI3K–AKT–mTOR signaling. Liver cancer is the second leading cause of cancer-related deaths worldwide, and hepatocellular carcinoma (HCC) accounts for approximately 90% of primary liver cancer cases. ![]() The book is frequently viewed as a metaphor for parenthood, with Victor being a negligent mother figure, abandoning his child - the Creature - after giving it life through his labors, and the troubled relationship that stems from his neglect. I mention that because that background bled into Frankenstein. Where the journeys of Lee & Clementine, Kratos & Atreus, and Joel & Ellie were exercises in bonding, this new crop is about exploring the complexities that stem from feelings of abandonment, uncertainty, absence - balanced alongside the more positive associations of family: safety, familiarity, assurance.įor context, Wollstonecraft passed away as a result of complications from childbirth after 11 days, and young Mary grew up the daughter of a distant stepmother while idolizing the legacy and work of her biological mother. They each speak to shared histories, which enables the characterization of the relationships to run deeper than those explored in the likes of Telltale’s The Walking Dead or God of War. Polly feels duty bound to find Ursula, even while she knows that finding her will be a bittersweet occasion due to their fraught relationship.Īcross these three stories, then, we see three different types of parent-child dynamics. ![]() For much of the game, that search is the driving force, and it’s bound up in complexity. No one knows where she went, so Polly’s quest to find her is based around investigation in both the “real” world and mythical realm of Reverie. The disappearance of Polly’s mother, Ursula, is unlike those of the two mentioned above in that it’s a complete mystery. Again, the missing mother is the inciting incident, resulting in protagonist Polly returning to her hometown and finding it not as she remembers. The other game that inspired these reflections is Don’t Nod’s latest, Harmony: The Fall of Reverie. ![]() There’s none of the teething issues or problematic intergenerational relationships that are so bound up in sad dad games. This single example shows a shift in the contexts of parenthood. In this, the game seems to apply a familiar sentiment of motherhood, that of protector, of salve, of someone you can rely on, even if only in memoriam. Venba’s missing mother is present in the game through a battered recipe book, which she turns to for comfort in a time of crisis. There’s no support network for her, and so, in a time of difficulty, she leans on the only thing she has left. The protagonist, Venba, is struggling to build a new life in a new country. One of them, Venba by Visai Games, was just a demo, so it’s hard to say the influence that element will have across the full game when it launches next week. In the past few weeks, I’ve come across three games that remove the sad dad from the center of the experience and place the missing mother there instead. But I wonder if their decline makes way for the rise of something both similar and different. As great as Telltale Games’ The Walking Dead, The Last of Us, and God of War are, they’re quite limited in how they use journey narratives and mostly conventional gameplay (or gameplay that has since become conventional) to explore the theme of parenthood. With the possible exception of the Max Payne remakes, I can’t think of a single major “sad dad” game on the horizon. Topias “Topson” Taavitsainen is no exception, and he is currently playing in the mid-lane position for his team. Tired of Dota 2 players? A fair chunk of that has gone to Ceb, and while some of his teammates have lost the ambition to compete, he has stuck around looking forward to the biggest TI event on record – 2020 TI. Sebastien “Ceb” Debs is one of OG’s own men who helped the team secure a back-to-back victory in the 20 International events, carrying his team to victory, which fetched OG $26,000,000 in total earnings from both events. However, ana’s hard work has paid off, and he has been able to win major tournaments in Dota 2 and become the third best-paid cyber athlete in the world.
